As soon as I read the title of this thread I instantly thought Monkey Island!!!! as the best PC game ever!
Best game EVER (regardless of format) has to be either:
Manic Miner
Secret of Mana (SNES) - Just amazing!
Very difficult question as everyone has their own opinion,
Personally for me I have too many, and in fact I cannot believe that Sensible Soccer on the Amiga wasnt mentioned!! Oh the zip sticks that I broke playing that game must run into the tens!!

When I fisrt got my 386 all i could play was stunt island it was addictive
also does anyone remember Pilot wings on the Famicon that was just great!! hours of fun sky diving thru rings of stars!!

Worms series, especially Armageddon
Lemmings
All Lucas Arts point and click really (Sam and Max, DOTT, Full Throttle...)
Shogun Total War
Unreal Tournament series...especially the first one
and my personal fave (and I still have it installed)...Virtual Pool 3 (awesome physics)

grand turismo 4 gets my vote excellent game
I agree with Welsh Pete you can't beat "worms armaggedon" I am still playing it on line with my family and freinds. Aslo any point and click game, I have just finished Runaway. Great game.
I came to post in favor of the Hitman series partly because of the replay value. Since its open ended you can complete your objects in countless ways, stealth or all out. Its one of the few games I have not grown tired of soon after completing.
To each his own they say. I know I absolutely cannot wait until the next one. ^_^
I cannot possibly believe that such avid gamers have not put X-COM on their list!
The defining moment in gaming for me were the first 3 hours on X-COM Terror from the Deep! :O

Wow, this is a hard one. I'm trying to judge it on games I've gone back to and can still play to this day.
Track 'n' Field (loved it in the arcade, praise to MAME for bringing it back. 24 years old I think this is now, and still playable.)
Bomb Jack (another MAME reinvention, at one point I held the third highest recorded score in the World for this game!)
Kick Off 2 (Amiga, preferred this to Sensi, maybe because it was out first and I played it more)
Zelda: Ocarina of Time (possibly the best game ever?)
Pro Evolution Soccer (the series - there is simply nothing like it in 2p mode, just nothing)
Tiger Woods (again the series - I reckon I've spent months on the virtual golf course over the years with this series )
Command & Conquer, Red Alert 2 for me I think.
